Is there a way to edit the templates to sort entries in the main index by karma? Or if not, perhaps a simple code modification?

Not that I know of. The code didn't seem simple either. However, Gm can do this with comments (see the tag {{logentrylist}} in gm_manual.html).
The method gm_generateentryloglistcomments is probably a good template. If it could be modified to work off of karma instead of comment number, it would probably suit your needs.
